Three things gang up on the second floor. Warm air rises, so heat from the whole house collects upstairs; that is the stack effect. The attic above the second-floor ceiling hits 150 degrees and radiates down through insulation that was adequate on paper. And the ductwork feeding the upstairs runs through that same attic, losing cooling to the heat around it before the air reaches a register. A single-zone system with one thermostat downstairs cannot see any of this. It satisfies the hallway and shuts off. Probably not. It is reading the hallway it lives in accurately; it simply cannot see the second floor. The fix is giving the upstairs its own sensor or thermostat, through zoning or a second system, so the equipment responds to the floor that is actually hot.
Closing one or two partially can help a little. Closing many raises static pressure, strains the blower and can freeze the coil, which trades a hot upstairs for no cooling at all. A measured balance at the trunk dampers does the same job without the side effects, and we test airflow before adjusting anything.
Sometimes, particularly in larger homes where one system was never enough for both floors. Often an upstairs return or a zoning retrofit gets there for far less. An on-site airflow test tells us which, which is why we do not recommend equipment from a phone call.
Frequently, if the duct layout allows dampers at the branch to each floor and the blower can handle the pressure when one zone closes. Single-stage systems usually need a bypass or careful damper sizing; variable-speed systems zone more gracefully. Our zoning page describes the retrofit.

Stack effect works in both directions. Warm air rises in summer and collects upstairs; in winter the same rising air leaves the downstairs cold while the upstairs is fine. Seasonal balancing, or zoning that adapts automatically, handles both.
